I have here investigated sub-spine actin organizations using<br />two-photon photoactivation of PA-GFP fused with β-actin in rat CA1 pyramidal<br />neurons in slice culture preparations. I found segregated and discontinuous<br /.>organizations of two pools of F-actin, dynamic and stable pools, which tuned<br />over with time constants of 1.2 min and 17 min, respectively. The stable F-actin<br />pool was localized at the base of spine head, often intruding into thick spine neck,<br />while the dynamic pool occupied the rest of spine head. Fractions of the stable<br />F-actin pool were greater in larger spines, therefore, the entire F-actin poor was<br />more stable in larger spines. I succeeded in visualizing a retrograde flow of<br />F-actin in the dynamic pool from the apex to the base of spine, and found that<br />both the speeds (0.2-1.2 μm/min) and lengths (0.2-0.7μm) of the F-actin flow<br />were greater in spines with larger head volumes.
