I examined expression patterns of the IL‐6 family members and their receptors during the rodent embryonic development,and found a specific expression of CNTF in the pineal gland and eyes. In vitro assays showed that newborn rat pineal extract supports some specific neurotransmitter/neuropeptide gene expressions[including substance P(SP)and vasoactive intestinal polypeptide(VIP);the gene expression of these peptides is known to be induced by CNTF in vitro] and survival of newborn rat sympathetic neurons. In contrast to the in vitro results,sympathetic innervation of the pineal gland in CNTF‐deficient mice showed no apparent difference from that of wild‐type mice.There were few preprotachykinin‐A(SP precursor protein)and VIP‐positive neurons among sympathetic neurons that innervate a pineal gland. These few VIP‐positive neurons in superior cervical ganglion were not different in number between adult CNTF knockout and wild‐type mice,implying that the CNTF signal in pineal glands may be usually unavailable for innervating sympathetic neurons.
